Quantification of color

First of all, in order to pursue a more objective evaluation, it uses a "metering device" as a measurement reference, and the picture taken by a microscope camera (microscope camera) is composed of red, green, and blue information. Then the measurement of color is actually the measurement of the red, green and blue information contained in the picture. The "measurement device" to test the color information of the picture is called Vectorscope, and the machine used by the team is the one on the right.

The screen has a circle with some small frames and lines. This circle corresponds to the familiar color wheel. The center of the color wheel is pure white, and the color saturation is higher toward the edge until it reaches the maximum saturation. If you send a picture to a vectorscope, the vectorscope will mark the color information of the picture on the screen. For example, if we want to measure whether the red shot by a microscope camera is real red, let the camera shoot a "standard red", and then send the picture to the vector oscilloscope. If the measurement result is marked in red, then this means The performance of the camera on red is accurate. If it is not in the red position, it is a color cast. To the left is yellow, and to the right is magenta (magenta).

So what color is "standard red"? Standard color is a color card issued by the generally recognized "color agency". In the field of microscopic imaging, there are similar institutions that will also release "standard colors" for testing. The more common microscope camera color cards include DSC target, Accu-Chart target, etc. The picture below (left) is a test target released by DSC Lab. There is a circle of color blocks arranged according to the order of the color wheel on this target board, among which there are standard red, green, blue, yellow, and magenta, and there are also intermediate transition colors; .

Use the microscope camera to shoot the test target, and then transfer the picture to the vector oscilloscope, and then check the standard position of each color to see whether the color of the CCD camera developed by it is accurate. This is how the professional R & D team tests the color accuracy of the imaging system.

Quantification of white balance

In the field of microscope imaging, another function that users often use is white balance. Similarly, the R & D team also needs to first standard white, white also has a standard white card, let the microscope camera adjust the white balance according to the standard white card and shoot a full white screen, and then observe the image of the vector oscilloscope in the computer. The vector oscilloscope image corresponding to pure white is a dot in the center of the screen. If it is not a dot, there is a color cast, whichever color is biased. For example, the test result on the right shows that the image is not all in the center, but the whole is shifted toward the red direction, and some are shifted toward the blue. This shows that the camera is slightly reddish and a little blued, and the color accuracy is clear at a glance.

It is more objective and true to evaluate the color performance of the camera in this way. We are using these measurement results to compare with other microscope cameras for true reduction.
